Understanding Vitamin K2 Forms

Naturally occurring vitamin K exists in two primary forms: K1 (phylloquinone), derived from green leafy vegetables and concentrated primarily in the liver, and K2 (menaquinones), synthesized by bacteria and found in animal-based and fermented foods. Among menaquinones, the most clinically relevant forms are MK-4 and MK-7 — with MK-7 demonstrating significantly superior pharmacokinetic properties.

MK-7’s longer aliphatic side chain makes it highly hydrophobic — a property that dramatically improves its transportability by plasma lipoproteins and its bioavailability in extrahepatic tissues (bones, arteries, and other peripheral tissues) where its activity is most needed. In contrast to K1, which is rapidly cleared by the liver, MK-7 circulates in plasma for days, supporting sustained carboxylation of K2-dependent proteins throughout the body.*

Osteocalcin and Bone Mineralization Osteocalcin is a protein produced by osteoblasts — the bone-building cells responsible for depositing new bone mineral. In its inactive, undercarboxylated form, osteocalcin cannot bind calcium ions effectively. Vitamin K2-dependent gamma-carboxylation activates osteocalcin, enabling it to bind calcium and incorporate it into the hydroxyapatite crystal structure of bone — the process that determines bone mineral density and bone quality. Adequate vitamin K2 status is essential for this carboxylation step, regardless of calcium or vitamin D3 intake.*

Matrix GLA Protein (MGP) and Cardiovascular Health Matrix GLA Protein (MGP) is a vitamin K2-dependent protein found in blood vessel walls and soft tissues throughout the body. In its active, carboxylated form, MGP is the body’s most potent known inhibitor of vascular and soft tissue calcification — it actively prevents calcium from depositing in arterial walls and other non-skeletal tissues. Without adequate vitamin K2 to carboxylate MGP, vascular calcium deposition is unsupported — a significant concern given the increasing awareness of the relationship between vascular calcification and cardiovascular health.*

Research has demonstrated that populations with higher menaquinone intake have higher rates of carboxylated MGP — reflecting adequate vitamin K2 activity in vascular tissue.*
